The film opens with Odin, King of Asgard, narrating the ancient war between the Asgardians and the Frost Giants of Jotunheim. The Asgardians emerged victorious, and Odin took the Casket of Ancient Winters from the Frost Giants' leader, Laufey. He also adopted an infant Frost Giant, whom he named Loki, and raised him alongside his biological son, Thor.
Centuries later, Thor, a powerful but arrogant warrior, is on the verge of being crowned King of Asgard. However, his coronation ceremony is interrupted when Frost Giants infiltrate Asgard's vault to retrieve the Casket. Thor, against Odin's express orders, leads his brother Loki, his childhood friend Sif, and the Warriors Three (Volstagg, Fandral, and Hogun) on a retaliatory attack to Jotunheim. Their impulsive assault nearly reignites the war between the realms. Odin intervenes, rescuing them, but is furious at Thor's arrogance and recklessness. As punishment, Odin strips Thor of his powers, banishes him to Earth (Midgard), and places an enchantment on his hammer Mjolnir: "Whosoever holds this hammer, if he be worthy, shall possess the power of Thor."

He attempts to retrieve Mjolnir, which has landed nearby and is now surrounded by a S.H.I.E.L.D. containment unit led by Agent Phil Coulson. Thor fails to lift the hammer, confirming Odin's enchantment, and is briefly captured by S.H.I.E.L.D. before Selvig secures his release. As he spends time with Jane, Thor begins to develop humility, compassion, and an understanding of humanity. A romance slowly blossoms between them.
Meanwhile, back on Asgard, Loki discovers a shocking truth about his lineage: he is actually Laufey's son, adopted by Odin after the war. When Odin falls into a deep "Odinsleep" to recover his strength, Loki seizes the opportunity to become acting King. He manipulates events, first allowing Laufey and the Frost Giants to infiltrate Asgard to kill Odin (which Loki secretly foils to appear heroic), and then sending the Destroyer, a powerful Asgardian automaton, to Earth to kill Thor and his new human friends.

As the Bifrost shatters, Loki allows himself to fall into the abyss of space, seemingly perishing. Odin awakens from his slumber and reconciles with Thor, who admits he is not ready to be king. Thor watches over Asgard, mourning Loki and longing for Jane. On Earth, Jane continues her research, determined to find a way to reach Thor again.
In a post-credits scene, Nick Fury takes Dr. Selvig to a S.H.I.E.L.D. facility and shows him the Tesseract, a mysterious cosmic cube of immense power. An unseen Loki subtly influences Selvig to agree to study it, setting up the events for The Avengers.
